<strong>Figure 3 Immunohistochemistry Validation of TRIM21 in Human Small Intestine Tissue </strong><br> Immunohistochemical analysis of paraffin-embedded human small intestine tissue using anti-TRIM21 antibody (7729) at 5 μg/ml. Tissue was fixed with formaldehyde and blocked with 10% serum for 1 h at RT; antigen retrieval was by heat mediation with a citrate buffer (pH6). Samples were incubated with primary antibody overnight at 4°C. A goat anti-rabbit IgG H&L (HRP) at 1/250 was used as secondary. Counter stained with Hematoxylin.

Independent Antibody Validation in Cell lines (Figure 2) shows similar TRIM21 expression profile in human cell lines detected by two independent anti-TRIM21 antibodies that recognize different epitopes, 7729 against central domain and 18-413 against recombinant fusion protein.  TRIM21 proteins are detected in the most tested cell lines at different expression levels by the two independent antibodies. 

Isoforms:
Human TRIM21 has 2 isoforms, including isoform 1 (475aa, 54kD) and isoform 2 (398aa, 45kD). Mouse TRIM21 has one isoform (470aa, 54kD) and Rat TRIM21 also has one isoform (471aa, 54kD). 7729 can detect human and possibly mouse and rat.

Background:
TRIM21, a member of the tripartite motif (TRIM) family, is a part of the RoSSA ribonucleoprotein, which includes a single polypeptide and one of four small RNA molecules. The RoSSA particle localizes to both the cytoplasm and the nucleus (1). RoSSA interacts with autoantigens in patients with Sjogren syndrome and systemic lupus erythematosus (2). TRIM21 also acts as a cytosolic antibody receptor; detection of intracellular antibodies activate immune signaling by stimulating the transcription factor pathways of NF-kappaB, AP-1, IRF3, IRF5, and IRF7, resulting in the production of proinflammatory cytokines and the induction of an antiviral state (3).
