To detect mRANTES by sandwich ELISA (using 100 ul/well antibody solution) a concentration of 0.5 - 2.0 ug/ml of this antibody is required. This antigen affinity purified antibody, in conjunction with ProSci’s Biotinylated Anti-Murine RANTES (XP-5259Bt) as a detection antibody, allows the detection of at least 0.2 - 0.4 ng/well of recombinant mRANTES.
To detect mRANTES by Western Blot analysis this antibody can be used at a concentration of 0.1-0.2 ug/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant mRANTES is 1.5-3.0 ng/lane, under either reducing or non-reducing conditions.

This antibody stained colchicine injected mouse brain (somatosensory cortex) tissue. The primary antibody was incubated at 0.5 ug/ml overnight at 4C. This was followed by a peroxidase conjugated secondary antibody and then a fluorescein Tyramide Signal Amplification (TSA) reagent. Optimal concentrations and conditions may vary.

Background:
RANTES is a chemoattractant for blood monocytes, memory T helper cells and eosinophils. It causes the release of histamine from basophils and activates eosinophils. It binds to CCR1, CCR3, CCR4 and CCR5 and is one of the major HIV suppressive factors produced by CD8+ T cells. Recombinant RANTES protein induces a dose dependent inhibition of different strains of HIV 1, HIV 2, and simian immunodeficiency virus (SIV). It is T cell and macrophage specific, induced by mitogens, and belongs to the intercrine beta (chemokine CC) family.
