Confocal imaging of NK-92 cells using GZMA Rabbit mAb followed by a further incubation with Cy3 Goat Anti-Rabbit IgG (H+L) (Red). DAPI was used for nuclear staining (Blue). Objective: 100x.
Immunoprecipitation of GZMA from 300 µg extracts of NK-92 cells was performed using 0.5 µg of GZMA Rabbit mAb. Rabbit IgG isotype control was used to precipitate the Control IgG sample. IP samples were eluted with 1X non-reducing Laemmli Buffer. The Input lane represents 10% of the total input. Western blot analysis of immunoprecipitates was conducted using GZMA Rabbit mAb at a dilution of 1:4000.

Background:
Cytolytic T lymphocytes (CTL) and natural killer (NK) cells share the remarkable ability to recognize, bind, and lyse specific target cells. They are thought to protect their host by lysing cells bearing on their surface 'nonself' antigens, usually peptides or proteins resulting from infection by intracellular pathogens. The protein described here is a T cell- and natural killer cell-specific serine protease that may function as a common component necessary for lysis of target cells by cytotoxic T lymphocytes and natural killer cells.
