<strong>Figure 6 Immunohistochemistry Validation of ZBP1 in Human Tonsil</strong><br>Immunohistochemical analysis of paraffin-embedded human tonsil tissue using anti-ZBP1 antibody (4401) at 1 μg/mL. Tissue was fixed with formaldehyde and blocked with 10% serum for 1 h at RT; antigen retrieval was by heat mediation with a citrate buffer (pH6). Samples were incubated with primary antibody overnight at 4°C. A goat anti-rabbit IgG H&L (HRP) at 1/250 was used as secondary. Counter stained with Hematoxylin.

Background:
Z-DNA Binding Protein 1 (ZBP1), or DNA-dependent activator of IFN-regulatory factors (DAI), is a pivotal protein in innate immunity and stress response. With two Z-DNA binding domains, ZBP1 senses cytosolic nucleic acids, triggering type I interferon responses. Beyond immunity, ZBP1 participates in necroptosis, a regulated cell death pathway. Dysregulation of ZBP1 is linked to autoimmune disorders and viral infections, making it a key player in disease pathogenesis.
